目的:研究脂氧素A4(lipoxinA4,LXA4)对子痫前期患者脐静脉内皮细胞分泌内皮素-1(endothelin-1,ET-1)的影响。方法:取子痫前期患者和正常孕妇组脐带及脐静脉血清,分离并培养脐静脉内皮细胞(human umbilical veinendothelial cell,HUVEC),分为正常细胞对照组、子痫前期细胞组和LXA4药物干预组(1 nmol/L,10 nmol/L,100 nmol/L)。RT-PCR检测各组细胞ET-1mRNA水平;酶联免疫吸附试验检测各组细胞上清液中ET-1的分泌水平。结果:LXA4降低子痫前期患者HUVEC内ET-1mRNA基因表达水平(P<0.05),而对正常细胞组无影响(P>0.05);LXA4明显抑制子痫前期脐静脉内皮细胞分泌ET-1(P<0.01),对正常细胞亦无影响(P>0.05)。结论:LXA4抑制子痫前期脐静脉内皮细胞ET-1 mRNA表达及分泌,为临床应用LXA4治疗妊娠期高血压疾病提供重要的实验依据。
Objective: To investigate the effect of lipoxin A4 (LXA4) on the secretion of endothelin-1 (ET-1) in human umbilical vein endothelial cells in preeclampsia. Methods: Umbilical cord and umbilical vein sera were collected from patients with preeclampsia and normal pregnant women. Human umbilical veinendothelial cells (HUVEC) were isolated and cultured and divided into normal control group, preeclampsia group and LXA4 intervention group (1 nmol / L, 10 nmol / L, 100 nmol / L). The levels of ET-1 mRNA in each group were detected by RT-PCR. The secretion of ET-1 in the supernatant of each group was detected by enzyme linked immunosorbent assay (ELISA). Results: LXA4 decreased the expression of ET-1mRNA in HUVECs (P <0.05), but had no effect on normal cells (P> 0.05). LXA4 significantly inhibited the ET-1 secretion in endothelial cells of preeclampsia P <0.01), had no effect on normal cells (P> 0.05). CONCLUSION: LXA4 can inhibit the expression and secretion of ET-1 mRNA in preeclampsia human umbilical vein endothelial cells and provide an important experimental basis for the clinical application of LXA4 in the treatment of hypertensive disorder complicating pregnancy.
