利用双微电极电压钳制技术研究丹参注射液(以下简称丹参)对离体家兔浦肯野纤维慢内向电流(Isi)的电生理作用。结果表明。丹参在浓度为1.0mg/ml 和2.0mg/ml,灌流20 min 时,能明显抑制 Isi,由给药前的76.25±20.83nA 降至53.13±18.70nA 和36.00±10.84 nA(P<0.05和0.01),对Isi 峰值抑制率分别为31%和53%,呈浓度依赖性抑制作用;在浓度为0.5mg/ml,刺激频率为1.0Hz,随着刺激教增加,丹参在开始抑制 Isi 的峰值不明显,以后抑制逐渐加强,直至稳态,呈使用依赖性阻滞作用。以上表明丹参具有钙拮抗剂作用。
The electrophysiological effects of Danshen injection (hereinafter referred to as Salvia miltiorrhiza) on the slow inward current (Isi) of isolated Purkinje fibers in rabbits were studied using a double microelectrode voltage clamp technique. The results show. Salvia miltiorrhiza was significantly inhibited by Isi at concentrations of 1.0 mg/ml and 2.0 mg/ml and perfused for 20 min, and decreased from 76.25±20.83 nA before administration to 53.13±18.70 nA and 36.00±10.84 nA (P<0.05 and 0.01). The peak inhibitory rate of Isi was 31% and 53%, respectively, in a concentration-dependent inhibition effect; at a concentration of 0.5 mg/ml, the stimulation frequency was 1.0 Hz. With the increase in stimulation education, the peak of Isi inhibition at the beginning of Salvia miltiorrhiza Obviously, the inhibition gradually increased until steady state, indicating a use-dependent blockade. The above shows that Danshen has a calcium antagonist effect.
