目的:观察人低分化鼻咽癌裸鼠移植瘤的微血管形态。方法:将人低分化鼻咽癌细胞单克隆株( C N E2 Z H5)接种于裸鼠皮下,然后对荷瘤鼠进行 A B S血管灌注,扫描电镜观察;移植瘤组织用因子Ⅷ相关抗原免疫组化( S P)方法观察血管形态。结果:(1)肿瘤微血管构筑表现为外型不规则、窦状、囊腔状血管形成,血管分级不明显;(2)血管壁薄,血管腔窄,数量少,管壁仅有一层内皮细胞,缺乏平滑肌,基底膜薄或缺如,内皮细胞间联接不牢固、呈开放状态,出现裂隙。结论:人低分化鼻咽癌裸鼠移植瘤血管构筑与正常微血管比较差异有显著。
Objective: To observe the microvascular morphology of human xenografted nasopharyngeal carcinoma in nude mice. Methods: Human low differentiated nasopharyngeal carcinoma cell clone (C N E 2 ZH5) was inoculated subcutaneously in nude mice, and then the tumor-bearing mice were subjected to A B S vascular perfusion and scanning electron microscopy. Transplanted tumor tissues were treated with factor Ⅷ related antigen immunohistochemistry (S P) method to observe the vascular morphology. Results: (1) The appearance of tumor microvessel showed irregular appearance, sinusoid and cystic vascularization, and the blood vessel grade was not obvious. (2) The vascular wall was thin, the vascular lumen was narrow and the number was small. There was only one layer of endothelial cell , The lack of smooth muscle, basilar membrane is thin or absent, the connection between endothelial cells is not strong, was open, cracks appear. Conclusion: There is significant difference between the angiogenesis and normal microvascular in human poorly differentiated nasopharyngeal carcinoma xenografts.
