目的研究白蔹甲醇提取物不同极性萃取部位对骨髓瘤细胞SP20增殖及凋亡的影响。方法体外培养骨髓瘤SP20细胞,光学显微镜观察加入白蔹甲醇提取物不同萃取部位后的细胞形态的变化;MTT法检测白蔹甲醇提取物不同极性萃取部位对骨髓瘤细胞增殖的影响。结果光学显微镜观察到给药后骨髓瘤SP20细胞出现典型的细胞凋亡形态改变。药物作用48 h后,5-氟尿嘧啶组、氯仿组、乙醚组、乙酸乙酯组对细胞增殖均有抑制作用,与溶剂空白组比较,差异有统计学意义(P<0.05,P<0.01);正丁醇组抑制率最小,与溶剂空白组比较,差异无统计学意义(P>0.05)。白蔹甲醇提取物不同极性萃取部位对骨髓瘤细胞存活的抑制率,从大到小依次是:乙酸乙酯组>乙醚组>氯仿组。结论白蔹甲醇提取物对体外培养的骨髓瘤细胞的生长有明显的抑制作用,并促其凋亡。
Objective To study the effects of different polar extracts of methanol extract from Baiji on the proliferation and apoptosis of myeloma cell line SP20. Methods The myeloma SP20 cells were cultured in vitro. The morphology of myeloma cell line SP20 was observed by optical microscope. The cell morphology was detected by MTT assay. The effects of different polar extracts on the proliferation of myeloma cells were detected by MTT assay. Results The morphological changes of typical myeloma SP20 cells were observed under light microscope. After 48 h treatment, 5 - Fluorouracil group, chloroform group, ether group and ethyl acetate group inhibited the cell proliferation. Compared with the solvent control group, the difference was statistically significant (P <0.05, P <0.01). The n-butanol group had the lowest inhibitory rate, which was not significantly different from that of the solvent blank group (P> 0.05). The inhibitory rates of different polar extracts of Bletilla striata on the survival of myeloma cells were as follows: ethyl acetate group> ether group> chloroform group. Conclusions The methanol extract of Bletilla striata can obviously inhibit the growth of myeloma cells in vitro and promote its apoptosis.
