目的 探讨增龄对大鼠骨骼肌细胞葡萄糖转运蛋白 4(glucosetransporter 4,GLUT4 )的影响。 方法 SD实验大鼠分为 2组 :青年组 (3月龄 )和老年组 (2 4月龄 )各 10只。制备GLUT4 羧基端12肽的多克隆抗体 ,利用Western印迹法检测 2组大鼠骨骼肌细胞GLUT4 蛋白含量 ,并检测大鼠尾静脉血糖。 结果 老年组大鼠的血糖 (5 6± 0 5mmol/L)略高于青年组大鼠 (4 5± 0 5mmol/L) ,但差异无显著性 ;青年组大鼠骨骼肌细胞内GLUT4 相对含量为 10 9 6 2± 12 2 5 ,而老年组为 86 46± 8 2 5 ,差异有显著性 (P <0 0 5 )。 结论 增龄可引起大鼠骨骼肌细胞GLUT4 蛋白含量明显减少 ,使骨骼肌细胞对葡萄糖的转运发生障碍 ,这可能是老年人易产生胰岛素抵抗的机制之一。
Objective To investigate the effects of aging on glucosetransporter 4 (GLUT4) in rat skeletal muscle cells. Methods SD experimental rats were divided into two groups: 10 in the young group (3 months old) and the elderly group (24 months old). The polyclonal antibody of GLUT4 carboxyl terminal 12 peptide was prepared. The content of GLUT4 in skeletal muscle cells of two groups was detected by Western blotting, and the tail vein glucose was measured. Results The blood glucose (5 6 ± 0 5 mmol / L) in the aged group was slightly higher than that in the youth group (45 ± 0 5 mmol / L), but the difference was not significant. The relative content of GLUT4 in the skeletal muscle cells Was 10 9 6 2 ± 12 2 5, while the elderly group was 86 46 ± 8 2 5, the difference was significant (P 0 05). Conclusions Aging can cause a marked decrease of GLUT4 protein in skeletal muscle cells of rats and block the transport of glucose by skeletal muscle cells, which may be one of the mechanisms that lead to insulin resistance in the elderly.
