目的研究中药复方保肝宁对酒精性肝纤维化大鼠的治疗作用,并探讨其抗肝纤维化的作用机制。方法建立酒精致肝纤维化大鼠模型,观察保肝宁对模型动物转化生长因子β1(TGFβ1)、基质金属蛋白酶组织抑制因子1(TIMP-1)、透明质酸梅(HA)、层粘连蛋白(LN)、三型前胶原(PCⅢ)、四型胶原(CIV)的影响。结果保肝宁能显著降低肝纤维化大鼠血清中的TGFβ1、TIMP-1、HA、LN、PCⅢ、CIV的表达水平,减轻肝纤维化大鼠的纤维增生程度。结论保肝宁有拮抗酒精致大鼠肝纤维化的作用,其机制可能与抑制肝纤维化形成中两个重要因子TGFβ1和TIMP-1的异常表达有关。
Objective To study the therapeutic effect of Baoganning, a traditional Chinese medicine compound, on rats with alcoholic liver fibrosis and to explore its mechanism of action against hepatic fibrosis. Methods Alcohol-induced hepatic fibrosis was induced in rats. The effects of Baoganning on the expression of transforming growth factor β1 (TGFβ1), tissue inhibitor of metalloproteinase 1 (TIMP-1), HA, laminin (LN), type III procollagen (PCIII), type IV collagen (CIV). Results Baoganning can significantly reduce the expression of TGFβ1, TIMP-1, HA, LN, PCⅢ and CIV in the serum of rats with hepatic fibrosis and reduce the degree of fibrosis in rats with hepatic fibrosis. Conclusion Baoganning can antagonize alcohol-induced hepatic fibrosis in rats. The mechanism may be related to inhibiting the abnormal expression of TGFβ1 and TIMP-1, two important factors in the formation of hepatic fibrosis.
