目的:探讨FTY720抑制人乳腺癌MCF-7细胞和喉癌Hep-2细胞增殖作用。方法:将人乳腺癌MCF-7细胞和喉癌Hep-2细胞用不同浓度FTY720作用48 h,MTT法检测细胞活性,流式细胞术检测其对细胞周期及凋亡率的影响。结果:MTT结果显示,FTY720对乳腺癌MCF-7细胞和喉癌Hep-2细胞增殖均有抑制作用,而且对喉癌Hep-2细胞抑制作用的浓度依赖性高于对乳腺癌细胞的作用;流式细胞术检测结果显示,FTY720可将乳腺癌MCF-7细胞阻滞于G1期,将喉癌Hep-2细胞阻滞于G2/M期,并明显促进乳腺癌MCF-7细胞凋亡。结论:一定浓度的FTY720能明显抑制体外培养的乳腺癌和喉癌细胞增殖,调节其细胞周期,诱导乳腺癌MCF-7细胞凋亡。
Objective: To investigate the inhibitory effect of FTY720 on the proliferation of human breast cancer MCF-7 cells and laryngeal carcinoma Hep-2 cells. Methods: Human breast cancer cell line MCF-7 and laryngeal cancer cell line Hep-2 were treated with different concentrations of FTY720 for 48 hours. The cell viability was measured by MTT assay. The cell cycle and apoptosis rate were determined by flow cytometry. Results: The results of MTT showed that FTY720 could inhibit the proliferation of MCF-7 breast cancer cells and Hep-2 cancer cells in a dose-dependent manner and the inhibitory effect on the proliferation of Hep-2 cells was higher than that of the breast cancer cells. Flow cytometry showed that FTY720 could arrest the MCF-7 breast cancer cells in G1 phase and arrest the laryngeal cancer Hep-2 cells in G2 / M phase and significantly promote the apoptosis of MCF-7 breast cancer cells. Conclusion: A certain concentration of FTY720 can significantly inhibit the proliferation of breast cancer and laryngeal carcinoma cells in vitro, regulate the cell cycle and induce the apoptosis of breast cancer MCF-7 cells.
