目的观察Pitavastatin对血管内皮细胞VEGF、bFGF等因子表达的影响,探讨他汀类药物可能具有的独立于血脂调节作用以外的促血管新生作用。方法体外培养HUVEC,加入不同浓度的Pitavastatin后用蛋白印迹杂交法检测VEGF,bFGF,p-Akt及p-eNOS蛋白表达水平;镉还原Griess测定培养基中NO含量。结果在培养的HUVEC中加入Pitavastatin后,VEGF,bFGF,p-Akt及p-eNOS蛋白呈现高表达,培养基中NO含量明显增高。结论Pitavastatin能诱导内皮细胞VEGF,bFGF,p-Akt及p-eNOS蛋白的表达及增加NO含量,推测其可能通过VEGF-p-Akt-eNOS(NO)径路而发挥非调脂的作用。
Objective To observe the effect of Pitavastatin on the expression of VEGF and bFGF in vascular endothelial cells and to explore the possible angiogenic effects that statins may have independent of the regulation of blood lipid. Methods HUVECs were cultured in vitro. The expression of VEGF, bFGF, p-Akt and p-eNOS were detected by Western blotting after adding different concentrations of Pitavastatin. The contents of NO in culture media were determined by cadmium reduction Griess. Results The expression of VEGF, bFGF, p-Akt and p-eNOS protein was significantly increased after adding Pitavastatin to cultured HUVECs. The content of NO in culture medium was significantly increased. Conclusion Pitavastatin can induce the expression of VEGF, bFGF, p-Akt and p-eNOS in endothelial cells and increase the content of NO. It is speculated that Pitavastatin may play a non-lipid-lowering role through the VEGF-p-Akt-eNOS pathway.
