目的检测在卵巢上皮性癌与非卵巢癌中,精子蛋白17(SP17)、精子相关抗原9(SPAG9)、OY-TES-1这三种癌-睾丸抗原基因启动子区域去甲基化表达的差异,探讨其与卵巢上皮性癌临床病理分期的关系。方法收集2015年1月至2016年4月25例新鲜的卵巢上皮性癌组织标本,及25例新鲜的非卵巢癌组织标本,采用DNA酸性亚硫酸盐转化、PCR扩增及甲基化敏感性内切酶方法检测SP17、SPAG9、OY-TES-1在卵巢上皮性癌与非卵巢癌中Cp G岛的去甲基化表达。结果与非卵巢癌组织比较,卵巢上皮性癌组织中SP17、SPAG9、OY-TES-1基因启动子区存在高去甲基化表达(P均<0.01)。三种癌-睾丸抗原基因启动子区域去甲基化表达阳性率在卵巢上皮性癌不同年龄、组织学类型和分化程度中比较,差异无统计学意义(P均>0.05);SP17、SPAG9启动子区域去甲基化表达阳性率在有淋巴结转移者中高于无淋巴结转移者(P<0.01,P<0.05),在Ⅲ~Ⅳ期患者中高于Ⅰ~Ⅱ期患者(P均<0.05);而OY-TES-1启动子区域去甲基化表达阳性率仅在有淋巴结转移者中高于无淋巴结转移者(P<0.05)。结论 SP17、SPAG9、OY-TES-1基因启动子区域在卵巢上皮性癌中存在高去甲基化表达,且其去甲基化表达均与患者发病年龄、组织学类型、分化程度无关;部分与淋巴结转移、肿瘤分期相关。SP17、SPAG9、OY-TES-1或可成为卵巢上皮性癌免疫治疗的靶位点。
Objective To detect the expression of promoter region of three cancer-testis genes in ovarian epithelial carcinoma and non-ovarian cancer, such as sperm protein 17 (SP17), sperm-associated antigen 9 (SPAG9) and OY-TES- Differences in ovarian epithelial cancer to explore its relationship with the clinical stage. Methods Twenty-five fresh specimens of epithelial ovarian cancer and 25 fresh specimens of non-ovarian cancer were collected from January 2015 to April 2016. DNA acid sulfite transformation, PCR amplification and methylation sensitivity Endo-enzyme method was used to detect the demethylation of SP17, SPAG9 and OY-TES-1 in CpG island in epithelial ovarian cancer and non-ovarian cancer. Results Compared with non-ovarian cancer, the expression of promoter region of SP17, SPAG9 and OY-TES-1 in ovarian epithelial carcinoma was highly demethylated (all P <0.01). The positive rates of demethylation of three cancer-testis gene promoter regions in ovarian epithelial cancer at different ages, histological types and differentiation were not significantly different (all P> 0.05); SP17 and SPAG9 were activated The positive rate of demethylation in subregion was higher in patients with lymph node metastasis than those without lymph node metastasis (P <0.01, P <0.05), and higher in patients with stage Ⅲ ~ Ⅳ than in patients with stage Ⅰ ~ Ⅱ (all P <0.05). The positive rate of demethylation in OY-TES-1 promoter region was higher than that in patients without lymph node metastasis (P <0.05). Conclusions The promoter regions of SP17, SPAG9 and OY-TES-1 gene are highly demethylated in epithelial ovarian cancer, and the demethylation of SP17, SPAG9 and OY-TES-1 genes are not associated with the age of onset, histological type and degree of differentiation. And lymph node metastasis, tumor staging related. SP17, SPAG9, OY-TES-1 or may be a target site for immunotherapy of epithelial ovarian cancer.
