本文利用~3H-TdR掺入法研究了油酸对小鼠艾氏腹水癌细胞DNA合成的影响。实验结果表明:油酸浓度为0.2mg/ml时,对~3H-TdR掺入DNA有一定阻抑作用。油酸浓度为2.0mg/ml时,作用强度增加,与对照组比较,P<0.01。两种浓度的油酸均在0.5小时对~3H-TdR掺入DNA的抑制作用最强,抑制率分别为56.8及66.7%。随时间延长,低浓度油酸的后作用减弱,4小时的抑制率为26.3%,而高浓度油酸4小时的抑制率为58.1%。与阿糖胞苷等药物比较,对~3H-TdR掺入的抑制与阿糖胞苷相近,掺入曲线均呈缓慢递增型,作用强度为阿糖胞苷的88.6%。综上所述,油酸能够影响~3H-TdR掺入艾氏腹水癌细胞DNA合成,其掺入抑制具有浓度依赖性。一旦去除药物的抑制,DNA的合成可以逐渐恢复。
In this paper, ~ 3H-TdR incorporation was used to study the effect of oleic acid on DNA synthesis in mouse Ehrlich ascites carcinoma cells. The experimental results show that the oleic acid concentration of 0.2mg / ml, ~ 3H-TdR incorporation of DNA have a certain inhibitory effect. The concentration of oleic acid 2.0mg / ml, the intensity increased, compared with the control group, P <0.01. Both concentrations of oleic acid had the strongest inhibitory effect on ~ 3H-TdR incorporation into DNA at 0.5 hour with the inhibition rates of 56.8 and 66.7%, respectively. With the extension of time, the post-action of low concentration of oleic acid weakened, the inhibition rate of 4 hours was 26.3%, while the inhibition rate of high-concentration oleic acid 4 hours was 58.1%. Compared with cytarabine and other drugs, the inhibition of ~ 3H-TdR incorporation was similar to that of cytarabine, and the incorporation curve showed a gradual increase with an intensity of 88.6% of that of cytarabine. In summary, oleic acid can affect ~ 3H-TdR incorporation into Ehrlich ascites cancer cells DNA synthesis, its incorporation inhibition in a concentration-dependent manner. Once the drug is removed from the inhibition, DNA synthesis can be gradually restored.
