为了研究胎鼠肝脏发育过程中肝窦的形成与成熟,用窦内皮细胞表达的一种特异性抗原,SE-S,作为窦内皮细胞表型成熟的标志,通过免疫组织化学和免疫电镜来观察肝脏发育的不同时期该抗原的表达以及与先存大血管的关系。实验证明,在胎龄15天时,即可观察到散在的肝窦样结构呈现阳性反应,而表达第八因子相关抗原的大血管为阴性。随着肝脏的发育,表达SE-S抗原的肝窦在数量和长度上亦随之增加,并且与肝细胞直接或密切接触。结果提示,肝窦的形成与成熟似乎是随机发生的,可能始于胚胎发育的第15天或更早,与先存大血管无直接相关性;肝细胞可能调节窦内皮细胞的成熟(SE-S抗原的表达)。
In order to study the formation and maturation of hepatic sinusoids during fetal liver development, a specific antigen expressed by sinus endothelial cells, SE-S, was used as a marker of sinusoidal phenotype maturation by immunohistochemistry and immunoelectron microscopy The expression of this antigen in different stages of liver development and the relationship with preexisting macrovascular. Experiments show that at 15 days gestational age, interspersed sinusoidal structures can be observed in a positive reaction, and the expression of the eighth factor-related antigens large blood vessels were negative. As the liver develops, the sinusoids that express the SE-S antigen also increase in number and length, and are in direct or intimate contact with hepatocytes. The results suggest that the formation and maturation of hepatic sinusoids appear to occur randomly, probably starting at day 15 or earlier of embryonic development and not directly related to preexisting macrovascular; hepatocytes may regulate sinusoidal endothelial cell maturation (SE- S antigen expression).
