目的:探索高分辨率熔解曲线(HRM)法检测肿瘤细胞系PTEN基因突变中的应用。方法:采用HRM法初筛肿瘤细胞系PTEN的突变,测序验证HRM筛选结果,TA克隆测序进一步明确突变情况。结果:HRM初筛出Jurkat细胞PTEN基因存在突变,测序结果与HRM结果一致,TA克隆测序确定Jurkat细胞系PTEN基因为缺失突变。结论:HRM法可准确筛选肿瘤细胞系中PTEN基因突变,适于肿瘤细胞PTEN基因突变筛选。
Objective: To explore the application of high resolution melting curve (HRM) in the detection of PTEN gene mutation in tumor cell lines. Methods: The mutation of PTEN in tumor cell line was detected by HRM. The result of HRM screening was verified by sequencing. The TA mutation was further confirmed by TA cloning and sequencing. Results: The mutation of PTEN gene in Jurkat cells was detected by HRM. The sequencing results were consistent with those of HRM. The deletion of PTEN gene in Jurkat cell line was confirmed by TA cloning sequencing. Conclusion: The HRM method can accurately identify PTEN gene mutations in tumor cell lines and is suitable for the screening of PTEN gene mutations in tumor cells.
