采用黑穗醋栗“丰产薄皮”与“亮叶厚皮”品种,对茎尖组织进行离体培养。通过调整培养基中BAP含量,获得了每20d达4.7的幼苗分化指数,幼苗在半量的MS无激素培养基上暗处理1~3d,接种密度每瓶3苗,生根率达83.3%~96.0%;将生根苗移栽于装有蛭石的木箱中,成活率达97%,并成功地移栽于大田,试管苗生长旺盛。建立了黑穗醋栗实用化快繁技术程序。
Using black currant “high yield thin skin” and “bright leaf thick skin” varieties, the tip tissue culture in vitro. By adjusting the content of BAP in the medium, the differentiation index of seedlings reached 4.7 every 20 days. Seedlings were dark-treated for 1 to 3 days on semi-amount of MS hormone-free medium and seedling density was 83.3% to 96.0% The rooting seedlings were transplanted in wooden boxes with vermiculite, the survival rate was 97%, and successfully transplanted in the field, test tube seedlings grow vigorously. The establishment of black currant practical fast propagation technology program.
