目的 寻找对胆管癌具有诊断价值的特异性标志物。方法 应用SDS PAGE分析 41例恶性胆道病变(胆管癌 2 7例、胆囊癌 9例、壶腹癌 5例 )和 2 0例良性胆道病变病人胆管胆汁中蛋白成分的差异。应用糖蛋白定性试验分析差异蛋白是否为糖蛋白。采用制备好的HRP标记凝集素探针 (DSA HRP、WGA HRP、LCA HRP、ConA HRP)对分离出的差异糖蛋白的糖链结构进行初步分析。结果 胆管癌胆汁中存在 4种差异较明显的蛋白 ,相对分子质量分别为 :75× 10 3、6 3× 10 3、5 2× 10 3、2 7× 10 3。前 3种为糖蛋白 ,后一种为非糖蛋白。胆管癌胆汁中分离出的 3种差异糖蛋白中糖链结构的变化 ,主要表现为天线数和核心岩藻糖的明显增多 ,还可能伴有平分型GlcNAc增多。结论 胆汁中某些糖蛋白及其糖链结构的变化可能对鉴别胆道良恶性病变有一定价值
Objective To search for specific markers that have diagnostic value for cholangiocarcinoma. Methods SDS PAGE was used to analyze the differences in bile duct bile protein composition in 41 malignant biliary tract lesions (27 cholangiocarcinomas, 9 gallbladder carcinomas, 5 ampulla cases) and 20 benign biliary tract lesions. A glycoprotein qualitative test was used to analyze whether the differential protein is a glycoprotein. The glycosylated structure of the isolated differential glycoproteins was initially analyzed using prepared HRP-labeled lectin probes (DSA HRP, WGA HRP, LCA HRP, ConA HRP). RESULTS There were four distinct proteins in the cholangiocarcinoma bile. The relative molecular masses were: 75×10 3, 6 3×10 3, 5 2×10 3, and 2 7×10 3. The first three are glycoproteins and the latter are non-glycoproteins. The changes in the sugar chain structure among the three differential glycoproteins isolated from bile duct bile are mainly manifested by a significant increase in the number of antennas and core fucose, and may also be accompanied by an increase in bisecting GlcNAc. Conclusion The changes of some glycoproteins and their sugar chain structure in bile may have some value in distinguishing benign and malignant biliary diseases.
