目的:采用转染的细胞构建裸鼠移植瘤,研究ALDH1A2过表达对异种移植瘤的影响。方法:利用不同组别的LOVO细胞接种裸鼠构建移植瘤模型,观察成瘤时间、瘤体平均体积,平均瘤重,探讨ALDH1A2在体内对结肠癌的影响。结果:ALDH1A2组及ATRA组裸鼠成瘤时间较对照组有显著延长,瘤体体积较对照组显著减少,瘤重也较对照组显著减轻,差异有统计学意义(P<0.05)。结论:成功地建立了人结肠癌细胞裸鼠移植瘤模型,AL-DH1A2组裸鼠成瘤时间短,瘤重低,ALDH1A2过表达可抑制LOVO细胞的增殖。
OBJECTIVE: To construct nude mice xenografts with transfected cells to study the effect of ALDH1A2 overexpression on xenograft tumors. Methods: Nude mice were inoculated nude mice with different groups of LOVO cells to establish tumor model. The tumorigenic time, the mean tumor volume and the average tumor weight were observed to investigate the effect of ALDH1A2 on colon cancer in vivo. Results: The tumorigenic time of ALDH1A2 group and ATRA group was significantly longer than that of the control group. The tumor volume was significantly reduced compared with the control group, and the tumor weight was also significantly reduced compared with the control group (P <0.05). Conclusion: The human colon cancer xenograft model was successfully established in nude mice. AL-DH1A2 group had shorter tumorigenesis time and lower tumor weight. ALDH1A2 overexpression inhibited the proliferation of LOVO cells.
