目的:HSV 可引起多种疾病,阿昔洛韦(ACV) 治疗HSV 感染效果虽好,但易出现耐药毒株;新药Cidofovir 对ACV 耐药毒株效果好,毒性也大。HSV 疫苗临床实验效果也不好。为了解决这些问题,必须继续开发新药。虎杖是一种抗病毒范围广的中药,为了了解虎杖的抗病毒性能并开发利用,我们对虎杖乙酸乙酯萃取部分的抗HSV 药效进行了研究。方法:我们以ACV 为阳性对照,利用病毒细胞病变抑制及空斑减数实验观察了药效,并且用中效作用原理对药效进行了分析。结果:在HEp2 细胞系统中,ACV 对HSV1F 株直接杀灭、增殖的抑制及感染阻断的ED50 分别为13 .22 ,9 .09 和8 .71μg/ml;晶Ⅳ的相应数值分别为0 .09 ,0 .07 和0 .03μg/ml;晶Ⅳ的三个治疗指数(TI)分别为ACVTI的307 ,272 和609 倍。ACV 对HSV2 333 株直接杀灭、增殖抑制及感染阻断的ED50 分别为16 .45 ,10 .85 和18 .62μg/ml;晶Ⅳ的相应数值分别为0 .07 ,0 .09 和2 .29μg/ml;晶Ⅳ的三个TI分别为ACVTI的498 ,251 和17 倍。结论:晶Ⅳ抗HSV 药效比ACV 强,毒?
Aims: HSV can cause a variety of diseases. Aciclovir (ACV) is effective in treating HSV infection, but resistant strains are easy to appear. The new drug Cidofovir is effective and resistant to ACV-resistant strains. HSV vaccine clinical trials are not effective. In order to solve these problems, we must continue to develop new drugs. Polygonum cuspidatum is a kind of traditional Chinese medicine with a wide range of anti-virus. To understand the anti-virus properties of Polygonum cuspidatum and its development and utilization, we studied the anti-HSV efficacy of ethyl acetate extract of Polygonum. Methods: We used ACV as a positive control, using the cytopathic effect of viral cytopathic effect and plaque reduction experiments to observe the efficacy, and using the principle of efficiency effect on the efficacy were analyzed. Results: In the HEp-2 cell system, ACV directly killed HSV 1F strain, and the inhibition of proliferation and ED50 of infection were 13, respectively. 22, 9. 09 and 8. 71μg / ml; corresponding values of crystal Ⅳ are 0 respectively. 09, 0. 07 and 0. 03μg / ml; three treatment index of crystal Ⅳ (TI) 307, 272 and 609 times ACVTI respectively.
