利用壳多糖缓慢降解的特性,把它与阿霉素制成壳多糖阿霉素缓释药粒,植入家兔胫骨上端进行体内释放试验,并与静注相同剂量阿霉素的家兔进行比较。结果表明,该缓释药可使阿霉素在局部骨组织中维持较高浓度达8周以上,且骨组织中最高浓度是静注相同剂量阿霉素时骨含量的29倍,但血浆峰值浓度仅为静注组血浆峰值的1/13。因此,壳多糖与阿霉素制成的缓释药粒具有良好的缓释功能,该缓释药粒阿霉素的一次局部用量可超过静注一次用量的10余倍,能较好地满足骨肿瘤局部化疗用药量大、维持时间长、血液浓度低、全身反应小的用药要求。
