目的 :探讨氨酪酸 (GABA)与空肠上皮细胞分化增生之间的关系。 方法 :用免疫荧光及激光共聚焦显微技术 ,对GABA及谷氨酸脱羧酶 (GAD ,包括两种异构体GAD6 5及GAD6 7)在大鼠空肠上皮中的表达进行研究。另以麦胚凝聚素组织化学染色与GAD6 5免疫荧光结合进行双重染色 ,显示GAD6 5表达细胞的分布特征。同时 ,用增生细胞核抗原 (PCNA)免疫组化方法显示上皮细胞的增生带。 结果 :GABA及GAD6 5阳性细胞主要分布在空肠绒毛的顶端 ,空肠上皮细胞GAD6 7阴性。双重染色显示 ,杯状细胞不表达GAD6 5。PCNA免疫染色阳性细胞主要在隐窝的中下部。表明GABA及GAD6 5主要分布在空肠上皮的成熟带及功能带。 结论 :GABA系统可能与上皮细胞的成熟及分化有关。
OBJECTIVE: To investigate the relationship between ammonia butyrate (GABA) and the differentiation and proliferation of jejunal epithelial cells. Methods: The expression of GABA and glutamate decarboxylase (GAD), including two isoforms GAD6 5 and GAD6 7, in the rat jejunum epithelium was studied by immunofluorescence and confocal microscopy. Another wheat germ agglutinin staining with GAD6 5 immunofluorescence combined with double staining, showing the distribution characteristics of GAD6 5-expressing cells. At the same time, proliferating zone of epithelial cells was detected by immunohistochemistry with proliferating cell nuclear antigen (PCNA). Results: The positive cells of GABA and GAD6 5 mainly distributed in the tip of jejunum villi, while the jejunal epithelial cells were negative for GAD6 7. Double staining showed that goblet cells did not express GAD6 5. PCNA immunostaining positive cells mainly in the lower part of the crypt. It showed that GABA and GAD6 5 are mainly distributed in the mature and functional bands of jejunum epithelium. Conclusion: GABA system may be related to epithelial cell maturation and differentiation.
