目的:研究三物降压汤(白毛夏枯草、马蹄决明子、炒杜仲)对血压、淋巴因子激活的杀伤细胞(LAK细胞)的影响。方法:将8周龄雄性自发性高血压大鼠(SHR)治疗后随机分成3组。治疗组(n=18)用三物降压汤(每日1ml,每毫升中含三物各0.5g)食管内喂饲。阳性对照组用卡托普利(每日10mg/kg,n=6),阴性对照组用生理盐水(n=6)喂饲,持续30天。用四唑盐(MTT)快速比色计量法测定LAK细胞的增殖与活性,用连苯三酚自氧化法测定LAK细胞表达的超氧化物歧化酶(SOD)样物质活性当量,用鲁米诺依赖化学发光法测定LAK细胞的自由基清除效应,用生物测定法观察了自发性高血压大鼠胸主动脉环对硝酸甘油和乙酰胆碱(Ach)的舒张反应及LAK细胞与SOD标准品对自发性高血压大鼠胸主动脉环对乙酰胆碱舒张反应的影响。结果:经三物降压汤治疗后,自发性高血压大鼠的血压下降,LAK细胞增殖、活性及其所表达的SOD样物质和其自由基清除效应均明显增强,与对照组比P<0.05。经LAK细胞(来自经三物降压汤治疗后的自发性高血压大鼠)孵育后的胸主动脉环对乙酰胆碱的舒张反应均有不同程度的增加,SOD标准品与LAK细胞有相同效应。结论:三物降?
OBJECTIVE: To study the effects of three substances blood pressure decoction (Prunella villosa, cassia cassia, and eucommia ulmoides) on blood pressure and lymphokine-activated killer cells (LAK cells). Methods: Eight-week-old male spontaneously hypertensive rats (SHR) were randomly divided into 3 groups. The treatment group (n=18) was fed intragastrically with three substances blood pressure decoction (1 ml per day, 0.5 g each in three ml). The positive control group was captopril (10 mg/kg daily, n=6), and the negative control group was fed with saline (n=6) for 30 days. The proliferation and activity of LAK cells were measured by tetrazolium (MTT) rapid colorimetric assay. The activity equivalent of superoxide dismutase (SOD)-like substances expressed in LAK cells was measured by pyrophenol autoxidation method. The free radical scavenging effect of LAK cells was determined by chemiluminescence assay. The relaxation response of thoracic aorta rings to nitroglycerin and acetylcholine (Ach) in spontaneously hypertensive rats and the spontaneousity of LAK cells and SOD standards were observed by bioassay. Effects of thoracic aortic rings on the relaxation of acetylcholine in hypertensive rats . RESULTS: After treatment with Sanwu Jiangya Decoction, the blood pressure of spontaneously hypertensive rats decreased, the proliferation and activity of LAK cells, and the expression of SOD-like substances and their free radical scavenging effects were all significantly enhanced. Compared with the control group, P< 0.05. After incubation of LAK cells (spontaneously hypertensive rats treated with Sanwu Jiangya Decoction), the thoracic aortic rings had different degrees of increase in the relaxation response to acetylcholine, and SOD standards had the same effect as LAK cells.
