目的:比较羟基红花黄色素A(HSYA)灌胃、耳缘静脉注射和股骨头髓腔内注射3种不同给药方式对激素性股骨头缺血坏死模兔股骨头细胞ERK1/2、JNK、p38蛋白表达的影响。方法:50只成年新西兰大白兔随机分为正常对照组、模型对照组、灌胃组、耳缘静脉注射组、髓腔内注射组,每组10只。除正常对照组外,其余各组每周2次臀肌注射7.5mg/kg醋酸泼尼松龙,连续6周,建立激素性股骨头缺血坏死模型。造模成功后,正常对照组和模型对照组不作治疗,其余3组分别给予HSYA灌胃、耳缘静脉注射和股骨头髓腔内注射治疗,4周、8周后各组分别处死5只,取出患侧股骨头,电镜观察股骨头细胞的超微结构,Western Blot检测股骨头细胞内ERK1/2、JNK、p38蛋白的表达。结果:4周后,髓腔内注射组ERK1/2蛋白表达较模型对照组明显升高(P<0.05),JNK、p38蛋白表达较模型对照组明显降低(P<0.05);8周后,耳缘静脉注射组、髓腔内注射组ERK1/2蛋白表达较模型对照组明显升高(P<0.05),JNK、p38蛋白表达较模型对照组明显降低(P<0.05)。结论:HSYA3种不同给药途径中,股骨头髓腔内注射的方式优于其余两种,可较好发挥药物疗效。
OBJECTIVE: To compare the effects of hydroxysafflor yellow A (HSYA) orally, auricular vein injection and intramedullary femoral injection on the expression of ERK1 / 2, JNK , P38 protein expression. Methods: Fifty adult New Zealand white rabbits were randomly divided into normal control group, model control group, gavage group, auricle vein injection group and intramedullary injection group, with 10 rats in each group. Except for the normal control group, the other groups were given glucocorticoid 7.5 mg / kg prednisolone twice a week for 6 consecutive weeks to establish a model of steroid-induced ischemic necrosis of the femoral head. After successful modeling, the normal control group and the model control group were not treated, the other three groups were given HSYA gavage, ear vein injection and femoral head intramedullary injection treatment, 4 weeks, 8 weeks after the death of each group were 5, The ipsilateral femoral head was removed and the ultrastructure of femoral head cells were observed by electron microscope. The expression of ERK1 / 2, JNK and p38 protein in the femoral head cells was detected by Western Blot. Results: After 4 weeks, the protein expression of ERK1 / 2 in the intramedullary cavity group was significantly higher than that in the model control group (P <0.05), and the expression of JNK and p38 was significantly lower than that in the model control group (P <0.05) Compared with the model control group, the expression of ERK1 / 2 protein in the rabbits in the ear vein and intramedullary injection group was significantly increased (P <0.05), while the expression of JNK and p38 protein in the rabbits was significantly lower than that in the model control group (P <0.05). CONCLUSION: Among the three routes of HSYA administration, the intramedullary femoral head injection is better than the other two, which can exert better therapeutic effect.
