目的以Gli-1 siRNA转染胰腺癌细胞系PANC-1,研究其阻断Hh-Gli-1信号通路的作用效果,并观察其对靶细胞增殖及凋亡的影响。方法实验组以Gli-1 siRNA转染PANC-1细胞,转染后24h采用RT-PCR检测Gli-1表达水平的变化,MTT法测定细胞生长曲线观察细胞增殖的抑制情况,同时设立阴性对照组和空白对照组。结果相对于阴性对照组和空白对照组,Gli-1 siRNA可以明显减少Gli-1 mRNA的表达(P<0.01),抑制PANC-1细胞的增殖(P<0.01)。结论实验初步证明Gli-1 siRNA通过阻断Gli-1 mRNA的转录,阻止下游基因的表达,从而抑制PANC-1的增殖并诱导细胞凋亡。
Objective To investigate the effect of blocking Hh-Gli-1 signal pathway by Gli-1 siRNA transfection into pancreatic cancer cell line PANC-1, and to observe its effect on proliferation and apoptosis of target cells. Methods The experimental group was transfected with PANC-1 cells with Gli-1 siRNA. The expression of Gli-1 was detected by RT-PCR 24h after transfection. The cell growth curve was measured by MTT assay to observe the inhibition of cell proliferation. At the same time, a negative control group And blank control group. Results Compared with the negative control group and the blank control group, Gli-1 siRNA significantly reduced the expression of Gli-1 mRNA (P <0.01) and inhibited the proliferation of PANC-1 cells (P <0.01). Conclusions The experiment initially proved that Gli-1 siRNA can inhibit the proliferation of PANC-1 and induce apoptosis by blocking the transcription of Gli-1 mRNA and preventing the expression of downstream genes.
