目的:探讨转录因子C/EBP-β在人正常子宫内膜及早孕蜕膜中的表达及其变化规律。方法:采用免疫组化进行组织学定位,采用免疫印迹进行蛋白质定量,采用RT-PCR进行mRNA检测,在蛋白质水平和mRNA水平检测人正常月经周期子宫内膜及早孕蜕膜中转录因子C/EBP-β的表达变化。结果:C/EBP-β蛋白主要在人子宫内膜基质细胞核中表达,C/EBP-β蛋白及mRNA在增生期C/EBP-β仅有弱表达,分泌期显著增加(P<0.05),以分泌晚期表达最强,在早孕蜕膜组织中也有较强的表达(P<0.05)。结论:C/EBP-β在人正常子宫内膜分泌晚期及早孕蜕膜组织中呈现较强的表达,可能在胚胎着床及妊娠的维持中发挥着重要的调节作用。
Objective: To investigate the expression of C / EBP-β in human normal endometrium and early pregnancy decidua and its variation. Methods: Immunohistochemistry was used for histological localization. Protein quantification was performed by Western blotting. MRNA was detected by RT-PCR. The levels of transcription factor C / EBP in endometrium and decidua at normal menstrual cycle and at mRNA and protein levels -β expression changes. Results: C / EBP-βprotein was mainly expressed in the nucleus of endometrial stromal cells. C / EBP-βprotein and mRNA were only weakly expressed in proliferative phase C / EBP-β and significantly increased in secretory phase (P <0.05) The secretion of late expression strongest, decidual tissue in early pregnancy also have strong expression (P <0.05). CONCLUSION: C / EBP-β is highly expressed in the endometrial secretion of human normal and decidual tissue in early pregnancy, which may play an important regulatory role in embryo implantation and pregnancy.
