目的通过研究β-连环蛋白(β-catenin)与胃癌组织分化程度及转移的关系,探讨β-catenin在胃癌发生及其转移的病理过程中的作用机理。方法应用Western Blot技术及免疫组化方法分析β-catenin在非恶性胃部组织、胃癌原发灶及转移灶组织中的表达,并对其进行比较。结果 Western Blot技术发现β-catenin在胃癌和非恶性胃部组织之间在表达的定量方面没有显著差异。免疫组化技术发现在163例原发性胃癌组织标本中,有79例β-catenin在细胞膜或者细胞浆阳性表达。β-catenin在低分化胃癌中表达水平显著降低(P<0.01)。41例转移性胃癌组织标本中有2例表达β-catenin,与原发性胃癌组织相比,表达水平显著降低(P<0.01)。结论β-catenin在部分原发性胃癌中表达缺失,在低分化胃癌及转移中表达缺失更为常见。
Objective To investigate the role of β-catenin in the pathogenesis of gastric cancer and its metastasis by studying the relationship between β-catenin and the differentiation and metastasis of gastric cancer. Methods Western blot and immunohistochemistry were used to analyze the expression of β-catenin in non-malignant gastric tissues, primary gastric cancer and metastatic tissues. Results Western Blot showed no significant difference in the quantitative expression of β-catenin between gastric cancer and non-malignant gastric tissues. Immunohistochemistry found that in 163 cases of primary gastric cancer specimens, 79 cases of β-catenin positive expression in the cell membrane or cytoplasm. The expression of β-catenin in poorly differentiated gastric cancer was significantly decreased (P <0.01). The expression of β-catenin in 2 of 41 metastatic gastric cancer tissues was significantly lower than that in primary gastric cancer (P <0.01). Conclusion The expression of β-catenin is not observed in some primary gastric carcinomas, but it is more common in poorly differentiated gastric carcinomas and metastases.
