目的:观察健脾化痰汤含药血清对胰岛素抵抗3T3-L1脂肪细胞分泌脂联素、TNF-α和IL-6的影响,探讨其改善IR可能机制。方法:采用细胞培养技术,将3T3-L1小鼠前脂肪细胞诱导分化为成熟脂肪细胞,通过地塞米松诱导建立IR细胞模型,将脂肪细胞分为正常组(基础培养液加10%空白组血清培养正常脂肪细胞)、IR组(基础培养液加10%空白组血清培养IR脂肪细胞)、中药组(基础培养液加10%健脾化痰组血清培养IR脂肪细胞)和西药组(基础培养液加10%比格列酮组血清培养IR脂肪细胞),培养48 h,葡萄糖氧化酶法检测细胞培养上清液中葡萄糖浓度,ELISA法检测细胞培养上清液中脂联素、TNF-α和IL-6含量。结果:与正常组比较,IR组葡萄糖消耗量和脂联素含量显著降低(P<0.05),TNF-α和IL-6含量显著升高(P<0.05);与IR组比较,中药组和西药组葡萄糖消耗量和脂联素含量明显升高(P<0.05),TNF-α和IL-6含量显著降低(P<0.05)。结论:脂联素、TNF-α和IL-6参与IR发生,健脾化痰汤可能通过调节IR脂肪细胞炎症细胞因子分泌,改善IR。
Objective: To observe the effect of Jianpi Huatan decoction-containing serum on the secretion of adiponectin, TNF-α and IL-6 in insulin resistance 3T3-L1 adipocytes and to explore its possible mechanism of improving IR. Methods: The 3T3-L1 mouse preadipocytes were induced to differentiate into mature adipocytes by cell culture technique. IR cell models were induced by dexamethasone, and the adipocytes were divided into normal group (basal medium supplemented with 10% blank serum , Normal adipocytes (IR), IR group (IR group). IR group (IR group) And 10% of the adiponectin group were cultured with serum of adiponectin group for 48 h. Glucose oxidase method was used to detect the concentration of glucose in the cell culture supernatant. The contents of adiponectin, TNF-α And IL-6 content. Results: Compared with the normal group, the glucose consumption and adiponectin content in IR group were significantly decreased (P <0.05), and the levels of TNF-α and IL-6 in IR group were significantly increased (P <0.05) Western medicine group glucose consumption and adiponectin levels were significantly increased (P <0.05), TNF-α and IL-6 levels were significantly lower (P <0.05). Conclusion: Adiponectin, TNF-α and IL-6 are involved in IR. Jianpi Huatan Decoction may improve IR by regulating the secretion of inflammatory cytokines in IR adipocytes.
