目的研究凋亡抑制基因蛋白Bcl-2与促进凋亡基因蛋白Hrk、细胞周期素CyclinE基因蛋白在胃癌中的表达,探讨其在胃癌发生、发展中的生物学关系及意义。方法应用S-P方法,检测58例胃癌组织及正常胃黏膜组织基因蛋白Bcl-2与Hrk、CyclinE表达情况。结果 Bcl-2蛋白在胃癌组织中阳性表达38例,阳性率为65.5%,在癌旁组织中阳性表达14例,阳性率为24.1%,两组中Bcl-2蛋白阳性率比较,差异有统计学意义(χ2=20.077,P<0.05)。不同肿瘤分化程度、临床分期及有无淋巴转移胃癌患者中Bcl-2蛋白阳性率比较,差异均有统计学意义(P<0.01);不同年龄、肿瘤直径、Borrmann分型、浸润深度胃癌患者中Bcl-2蛋白阳性率比较,差异均无统计学意义。Hrk蛋白在癌旁组织中阳性表达36例,阳性率为62.1%,在胃癌组织中阳性表达12例,阳性率为20.7%,两组中Hrk蛋白阳性率比较,差异有统计学意义(χ2=20.471,P<0.05)。CyclinE蛋白在胃癌组织中阳性表达34例,阳性率为58.6%,在癌旁组织中阳性表达11例,阳性率为18.9%,两组中Hrk蛋白阳性率比较,差异有统计学意义(χ2=19.206,P<0.05)。Bcl-2蛋白与Hrk蛋白和CyclinE蛋白的相关分析,Bcl-2蛋白与Hrk蛋白呈负相关(rs=-0.312,P<0.05),与CyclinE蛋白呈正相关(rs=0.682,P<0.05)。结论 Bcl-2、Hrk及CyclinE基因蛋白均与胃癌发生有关,三者相互制约或协调,参与胃癌形成机制。
Objective To study the expression of Bcl-2, Hrk and CyclinE in gastric cancer and to explore their biological significance in the development and progression of gastric cancer. Methods S-P method was used to detect the expression of Bcl-2, Hrk and CyclinE in 58 cases of gastric cancer and normal gastric mucosa. Results The positive rate of Bcl-2 protein in gastric cancer tissues was 38 cases, the positive rate was 65.5%. The positive rate of Bcl-2 protein in paracancerous tissues was 14.1%. The positive rates of Bcl-2 protein in both groups were statistically different Significance (χ2 = 20.077, P <0.05). There were significant differences in the positive rate of Bcl-2 protein in different degree of tumor differentiation, clinical stage and lymph node metastasis of gastric cancer (P <0.01); in different age, tumor diameter, Borrmann type, depth of invasion of gastric cancer Bcl-2 protein positive rate comparison, the difference was not statistically significant. Hrk protein was positively expressed in adjacent tissues (36 cases), the positive rate was 62.1%. The positive rate of Hrk protein was 12.7% in gastric cancer tissues. The positive rate of Hrk protein in the two groups was statistically significant (χ2 = 20.471, P <0.05). The positive expression rate of CyclinE protein in gastric cancer tissues was 34 cases, the positive rate was 58.6%. The positive rate of CyclinE protein in adjacent tissues was 11.9% (positive rate was 18.9%). The positive rate of Hrk protein in the two groups was statistically significant (χ2 = 19.206, P <0.05). Bcl-2 protein was negatively correlated with Hrk protein (rs = -0.312, P <0.05), and positively correlated with CyclinE protein (rs = 0.682, P <0.05). Conclusion Bcl-2, Hrk and CyclinE gene proteins are all involved in gastric carcinogenesis. The three proteins are restricted or coordinated with each other and participate in the formation of gastric cancer.
