目的探讨黄连素对前列腺癌LNCaP细胞裸鼠移植瘤的放射增敏效应及作用机制。方法建立前列腺癌LNCaP细胞裸鼠移植瘤模型;移植瘤平均体积增至约125 mm3时,将裸鼠24只随机分为4组:对照组、单纯药物组(5 mg/kg黄连素)、单纯照射组(8 Gy)、药物+照射组(5 mg/kg黄连素+8 Gy)。36 d后测量移植瘤体积,计算肿瘤体积抑制率,并对移植瘤标本进行Western blot检测移植瘤组织中HIF-1α及VEGF蛋白的表达。结果药物+照射组裸鼠移植瘤体积为(505.0±110.7)mm3,显著低于单纯药物组(1384.2±182.0)mm3和单纯照射组(1119.7±248.2)mm3(P<0.05),其抑制率达(74.5±4.4)%。Western blot结果显示,药物+照射组移植瘤组织中HIF-1α、VEGF蛋白表达水平较单纯照射组明显降低(P均<0.0001)。结论黄连素对前列腺癌LNCaP细胞裸鼠移植瘤具有放射增敏作用,可能与其抑制HIF-1α、VEGF途径有关。
Objective To investigate the radiosensitizing effect of berberine on the transplanted tumor of prostate cancer LNCaP cells in nude mice and its mechanism. Methods The nude mice model of prostate cancer LNCaP was established. Twenty-four nude mice were randomly divided into 4 groups: control group, pure drug group (5 mg / kg berberine) Irradiation group (8 Gy), drug + irradiation group (5 mg / kg berberine +8 Gy). The volume of tumor xenografts was measured after 36 days. The tumor volume inhibition rate was calculated. The expression of HIF-1α and VEGF protein in xenografts was detected by Western blot. Results The tumor volume of nude mice in the drug + irradiation group was (505.0 ± 110.7) mm3, which was significantly lower than that in the pure drug group (1384.2 ± 182.0 mm3) and in the irradiation group (1119.7 ± 248.2) mm3 (P <0.05) (74.5 ± 4.4)%. Western blot results showed that the expression of HIF-1α and VEGF protein in the tumor + drug group was significantly lower than that in the radiation group (all P <0.0001). Conclusion Berberine radiosensitizes the transplanted tumor of prostate cancer LNCaP cells in nude mice, which may be related to the inhibition of HIF-1α and VEGF pathway.
