本实验用氨基糖甙类抗生素卡那霉素造成豚鼠中毒性耳聋,观察内耳听毛细胞、血管纹细胞超微结构的变化。结果见受损听毛细胞溶酶体增多、部分线粒体肿胀变性、板层小体增多、传出末梢突触囊泡减少;血管纹边缘细胞部分线粒体变性、糙面内质网减少。实验提示:听毛细胞及其底部的神经末梢、血管纹边缘细胞是卡那霉素耳毒作用的主要部位。药物可能通过干扰内耳细胞的自身代谢、能量代谢、蛋白质的合成以及神经递质的代谢而致聋。
In this experiment, the aminoglycoside antibiotic kanamycin caused detoxification in guinea pigs and observed the changes of the ultrastructure of inner ear hair cells and vascular leukocytes. The results showed that damaged hearing cells increased lysosomes, some mitochondria swelling degeneration, lamina propria increased, the amount of peripheral synaptic vesicles decreased; edge of the stria vascularis mitochondria degeneration, rough endoplasmic reticulum decreased. Experimental Tip: Listen to the bottom of the hair cells and the nerve endings, stria vascularis cells kanamycin ototoxicity is the main part. Drugs may cause deafness by interfering with the intrinsic metabolism of the inner ear cells, energy metabolism, protein synthesis, and metabolism of neurotransmitters.
