目的:观察潜阳育阴颗粒(Qianyang Yuyin granules,QYG)对正常麻醉犬肾脏微循环影响,并初步研究其作用机制。方法:应用Power Lab系统记录十二指肠给予潜阳育阴颗粒前和给药后15~180 min不同时段比格犬肾脏微循环变化;犬分为正常组,QYG高、中、低剂量组(12.30,6.15,3.08 g·kg-1),卡托普利组(30 mg·kg-1)和珍菊降压片组(0.14 g·kg-1),于给药前及给药后15,30,60,90,120,180 min通过股动脉取血,ELISA法检测血清中血管紧张素Ⅱ(AngⅡ),内皮型一氧化氮合酶(e NOS)含量,化学法检测一氧化氮(NO)含量;给药3 h后取肾脏组织,ELISA法检测肾脏AngⅡ含量。结果:潜阳育阴颗粒12.30 g·kg-1剂量组给药后60~180 min正常麻醉犬的肾动脉微循环血流量明显上升(P<0.05),给药180 min后肾脏组织AngⅡ含量明显下降(P<0.05),给药90,120,180 min后,血清中AngⅡ明显下降(P<0.05),e NOS和NO含量明显上升(P<0.05);潜阳育阴颗粒6.15 g·kg-1剂量组给药后90~120 min内正常麻醉犬的肾动脉微循环血流量明显上升(P<0.05),给药90,120 min后,血清中AngⅡ明显下降(P<0.05),e NOS和NO含量明显上升(P<0.05);潜阳育阴颗粒低剂量组给药后180min内正常麻醉犬各项指标变化不明显。结论:潜阳育阴颗粒可改善肾脏微循环,其作用机制可能与降低循环及肾脏组织AngⅡ的含量,增加循环中e NOS及NO含量有关。
Objective: To observe the effects of Qianyang Yuyin granules (QYG) on renal microcirculation in anesthetized dogs and to study its mechanism. Methods: The changes of renal microcirculation in beagle dogs were observed before and after administration of Qiangyouyu Yin granule by using Power Lab system. The dogs were divided into normal group, QYG high, medium and low dose groups (12.30,6.15,3.08 g · kg-1), captopril group (30 mg · kg-1) and Zhenju Xiajue Tablet group (0.14 g · kg-1) 15, 30, 60, 90, 120 and 180 min. Blood samples were collected from the femoral artery. Serum levels of angiotensin Ⅱ and eNOS were detected by ELISA. Nitric oxide (NO) After 3 h, the kidneys were taken out and the content of AngⅡ in kidneys was detected by ELISA. Results: The blood flow of microcirculation of renal artery in normal anesthetized dogs was significantly increased (P <0.05) at 60 ~ 180 min after administration of Qiangyou-Yin-Yin Granules in 12.30 g · kg-1 dose group (P <0.05). After 90,120 and 180 min of administration, the level of AngⅡ in serum decreased (P <0.05) and the content of eNOS and NO increased obviously (P <0.05) The blood flow of microcirculation of renal arteries in normal anesthetized dogs was significantly increased within 90-120 min after administration (P <0.05), while the levels of AngⅡ in serum were significantly decreased (P <0.05) 90 ng and 120 min after administration, and the content of eNOS and NO was significantly increased (P <0.05). The indexes of normal anesthetized dogs in sub-yang yin-yin low dose group did not change significantly within 180 minutes after administration. CONCLUSION: QianYangYuYin granules can improve renal microcirculation, and its mechanism may be related to reducing the content of Ang Ⅱ in circulation and kidney tissue and increasing the content of eNOS and NO in circulation.
