目的比较苦豆子总碱及8种生物碱单体,筛选出具有抗癌活性的药物。方法采用MTT方法测定苦豆子总碱及8种生物碱单体对肝癌、胃癌、结肠癌细胞株的增殖活性影响;Hochest33342-PI双染观察细胞形态学变化,流式细胞仪检测细胞周期及细胞凋亡。结果苦豆子总碱及8种生物碱对消化道癌细胞具有不同程度的增殖抑制作用,其作用呈时间-剂量依赖性,以苦豆碱的抑制作用最强。苦豆碱促进了细胞凋亡,并使细胞阻滞在G2期。结论苦豆子总碱及8种生物碱单体都具有一定的抗癌活性,以苦豆碱的活性最强,为进一步开发抗癌新药提供实验依据。
Objective To compare the total alkaloids of Sophora alopecuroides and eight kinds of alkaloids to screen out the anticancer drugs. Methods The MTT method was used to determine the effects of total alkaloids from Sophora alopecuroides and eight alkaloids on the proliferation of hepatoma, gastric cancer and colon cancer cell lines. Hochest33342-PI double staining was used to observe the cell morphological changes. The cell cycle and cell cycle were detected by flow cytometry Apoptosis. Results Total alkaloids of Sophora alopecuroides and eight kinds of alkaloids had different degrees of proliferation inhibitory effects on gastrointestinal cancer cells. The effects of time were dose-dependent and the inhibitory effects of paracetamol were the strongest. Alperitine promotes apoptosis and blocks cells in the G2 phase. Conclusion The total alkaloids of Sophora alopecuroides and eight kinds of alkaloids have some anticancer activities, and the activity of paracetamol is the strongest, which provides experimental basis for further development of new anticancer drugs.
