目的 探讨趋化因子RANTES在狼疮肾炎的表达情况及其对狼疮肾炎肾损伤的作用。方法 复制慢性移植物抗宿主(GVHD)小鼠模型,应用免疫组织化学和逆转录多聚合酶链式反应检测不同时期狼疮样小鼠肾组织RANTES的表达。结果(l)正常小鼠肾组织中未见RANTES表达,8周狼疮样小鼠肾组织中,见部分肾小管上皮细胞表达RANTES;12周狼疮样小鼠肾组织中,大部分肾小管表达强的RANTES,并与小管间质损伤总积分呈正相关关系(r=0.912,P<0.01),肾小球RANTES阳性细胞数与肾小球硬化指数也呈正相关关系(r=0.837,P<0.01);(2)正常小鼠肾组织中未见RANTES mRNA表达,12周狼疮样小鼠肾组织 RANTES mRNA表达明显高于8周狼疮样小鼠,RANTES mRNA/β-actin mRNA有显著性差异(P<0.05) 结论 RANTES在自身免疫性肾炎中起重要作用,是加剧肾损伤的关键因子之一.
Objective To investigate the expression of chemokine RANTES in lupus nephritis and its effect on renal injury induced by lupus nephritis. METHODS: Chronic graft versus host (GVHD) mouse models were duplicated and the expression of RANTES in renal tissue of lupus-like mice was detected by immunohistochemistry and reverse transcriptase-polymerase chain reaction. Results (1) The expression of RANTES was not found in the normal mouse kidney. In the 8 weeks, the renal tubular epithelial cells of the lupus-like mouse expressed RANTES. In the 12 weeks, the expression of most of the renal tubules in lupus-like mice (R = 0.912, P <0.01). There was also a positive correlation between the number of RANTES positive cells and glomerular sclerosis index (r = 0. 837, P <0.01). (2) The expression of RANTES mRNA in renal tissues of normal mice was not detected. The expression of RANTES mRNA in renal tissue of lupus-like mice in 12 weeks was significantly higher than that in 8 weeks (P <0.05) Conclusions RANTES plays an important role in autoimmune nephritis and is one of the key factors aggravating renal injury.
