目的 观察人参提取物在内毒素脂多糖 (LPS)所致的SIRS MODS中对脾细胞的作用。 方法 通过HE、TUNEL染色和透射电镜比较注射人参提取物 (GS)后再注射LPS时的脾脏形态学及凋亡情况 ,并用免疫组织化学的方法检测Bcl 2 ,Fas,FasL的表达。 结果 随LPS作用时间的延长 ,脾损伤逐渐加重。动脉周围淋巴鞘细胞碎片及TUNEL阳性细胞增多 ,电镜观察可见到凋亡的淋巴细胞。而人参组细胞损伤明显减轻。Bcl 2的表达随着LPS作用时间的延长而减少 ,Fas ,FasL则增强。人参提取物主要阻止Bcl 2表达减少。 结论 人参提取物对LPS脾细胞损伤有保护作用 ,其作用机制与增强Bcl 2表达有关
Objective To observe the effect of ginseng extract on spleen cells in SIRS MODS induced by endotoxin lipopolysaccharide (LPS). Methods Morphology and apoptosis of spleen were measured by HE, TUNEL staining and transmission electron microscopy after injection of ginseng extract (GS) followed by injection of LPS. The expression of Bcl 2 and Fas and FasL were detected by immunohistochemistry. Results With the prolongation of the LPS action time, splenic injury gradually increased. Peripheral lymphatic sheath cell debris and TUNEL-positive cells were increased. Electron microscopic observations revealed apoptotic lymphocytes. The ginseng group significantly reduced cell damage. The expression of Bcl 2 decreased with the prolongation of LPS, while Fas and FasL increased. Ginseng extracts mainly prevent the decrease of Bcl 2 expression. Conclusion Panax ginseng extract has protective effect on LPS spleen cell injury, and its mechanism of action is related to enhanced Bcl 2 expression.
