目的探讨2008年山东省流行的肠道病毒71型(EV71)的基因特征。方法以2008年山东省分离的一株EV71(JN200804)为样本,采用RT-PCR方法扩增并测定全基因组核苷酸序列,进行同源性比较和种系发生分析。结果 JN200804株的全基因组序列长度为7404个核苷酸,与其他EV71毒株相比,在编码区没有核苷酸的缺失和插入,5′UTR和3′UTR的长度和序列有差异。核苷酸同源性比较结果表明,JN200804株与安徽株(Fuyang/17.08/1)、浙江株(Zhejiang08)和北京株(BJ08)的核苷酸同源性均大于95%,与国际标准株BrCr的同源性仅为79.8%。氨基酸同源性比较结果表明,JN200800株与Fuyang/17.08/1同源性高达99.6%,与TW/2086/98同源性最低(94.5%)。根据P1、P2和P3区基因序列构建了种系进化树:JN200804株与C4亚型各毒株(Fuyang/17.08/1、Zhejiang08、BJ08、SHZH03、N3340-TW-02和SHZH98)同属一个分支,核苷酸同源性都在91%以上。结论 JN200804株属于EV71的C4亚型,与AnhuiFY08、Zhejiang08和BJ08的进化关系最为密切。
Objective To investigate the genetic characteristics of enterovirus 71 (EV71) in Shandong Province in 2008. Methods An EV71 (JN200804) isolated from Shandong province in 2008 was used as a sample. The nucleotide sequence of the whole genome was amplified and sequenced by RT-PCR. The homology comparison and phylogenetic analysis were performed. Results The genome sequence of JN200804 was 7404 nucleotides in length. Compared with other EV71 strains, there was no nucleotide deletion and insertion in the coding region. The length and sequence of 5’UTR and 3’UTR were different. Nucleotide homology comparison showed that nucleotide homology between JN200804 strain and Fuyang / 17.08 / 1 strain, Zhejiang08 strain and Beijing strain (BJ08) was greater than 95% The homology of BrCr is only 79.8%. Comparison of amino acid homology showed that the homology of JN200800 with Fuyang / 17.08 / 1 was as high as 99.6%, the lowest homology with TW / 2086/98 (94.5%). The phylogenetic tree was constructed based on the P1, P2 and P3 gene sequences. JN200804 strain belongs to the same branch as the C4 subtype strains (Fuyang / 17.08 / 1, Zhejiang08, BJ08, SHZH03, N3340-TW-02 and SHZH98) Nucleotide homology is above 91%. Conclusion The strain JN200804 belongs to the C4 subtype of EV71 and has the closest evolutionary relationship with AnhuiFY08, Zhejiang08 and BJ08.
