目的研究表皮生长因子(EGF)、神经降压素(NT)、降钙素基因相关肽(CGRP)对四氯化碳(CCl_4)损伤原代培养肝细胞的保护作用。方法建立大鼠离体培养肝细胞损伤模型,设对照组、损伤组及不同浓度胃肠肽预处理组(胃肠肽提前1h加入),24h后测上清液酶水平、肝细胞SOD活性和MDA(丙二醛)含量,以台盼蓝染色试验计算肝细胞存活率,观察形态学变化。结果 EGF、NT明显降低原代培养肝细胞CCl_4损伤后上清液酶学水平、MDA含量,提高SOD活性和细胞存活率(P<0.01),改善形态学变化。CGRP对上述指标和形态学改变无明显影响。结论 EGF、NT对离体培养肝细胞CCl_4损伤有直接保护作用,而CGRP无此作用。EGF、NT可能是通过稳定生物膜、抗脂质过氧化起作用的。
Objective To investigate the protective effect of epidermal growth factor (EGF), neurotensin (NT) and calcitonin gene related peptide (CGRP) on primary culture hepatocytes injured by carbon tetrachloride (CCl 4). Methods Rat liver injury model was established. The control group, injury group and different concentrations of gastrointestinal peptide pretreatment group (gastrointestinal peptide added 1h earlier), 24h after the supernatant enzyme level, SOD activity and MDA (malondialdehyde) content, trypan blue staining test calculated liver cell viability, observe the morphological changes. Results EGF and NT significantly decreased the level of serum and MDA, increased the activity of SOD and the survival rate of cells (P <0.01), and improved the morphological changes of supernatant after CCl_4 injury in primary cultured hepatocytes. CGRP had no significant effect on the above indexes and morphological changes. Conclusion EGF and NT have a direct protective effect on CCl 4 injury in cultured hepatocytes, but CGRP does not. EGF, NT may play a role in anti-lipid peroxidation by stabilizing biofilms.
