本研究旨在探索bag3基因在慢性淋巴细胞白血病(chronic lymphocytic leukemia,CLL)中的表达及与患者预后的关系。通过实时定量PCR技术,采用SYBR Green荧光染料法对46例CLL患者外周血标本bag3 mRNA表达水平进行相对定量检测,并分析CLL患者bag3基因异常表达和临床预后指标的相关性。结果表明,46例CLL患者bag3表达水平中位数为0.021(0.0007-1.124),明显高于正常人群[0.0025(0.0005-0.014)];耐药CLL患者的bag3表达水平明显高于治疗有效患者的bag3表达水平;bag3的表达与性别、年龄、淋巴细胞数、临床分期、IgVH基因突变、染色体异常、CD38、ZAP70无明显相关。结论:bag3基因在CLL中的表达水平明显高于正常人群,可能与白血病细胞耐药相关,而与CLL患者临床分期及临床常规预后因素无明显相关。
This study aimed to explore the relationship between the expression of bag3 gene and the prognosis of patients with chronic lymphocytic leukemia (CLL). Real-time quantitative PCR technique was used to detect the expression of bag3 mRNA in peripheral blood of 46 patients with CLL by SYBR Green fluorescent dye method. The correlation between the abnormal expression of bag3 gene and clinical prognostic factors was analyzed. The results showed that the median of bag3 expression in 46 patients with CLL was 0.021 (0.0007-1.124), which was significantly higher than that of normal people [0.0025 (0.0005-0.014)]; bag3 expression in patients with drug-resistant CLL was significantly higher than that in patients with active CLL bag3 expression; bag3 expression and gender, age, lymphocyte number, clinical stage, IgVH gene mutation, chromosomal abnormality, CD38, ZAP70 no significant correlation. Conclusion: The expression of bag3 gene in CLL is significantly higher than that in normal people, which may be related to the drug resistance of leukemia cells. However, there is no significant correlation between the expression of bag3 gene and clinical stage and clinical prognostic factors.
