目的:对六味地黄苷糖中糖苷部位的化学成分进行研究,阐明其活性的物质基础。方法:用大孔树脂柱色谱制备糖苷部位,用硅胶和反相硅胶柱色谱分离各成分,用MS及NMR技术鉴定结构。结果:分离得到了11个化合物,其中包括9个环烯醚萜苷,1个蒎烷单萜苷,1个酚苷,分别鉴定为7-脱氢马钱素(1),7α-甲氧基莫诺苷(2),7β-甲氧基莫诺苷(3),7α-乙氧基莫诺苷(4),7β-乙氧基莫诺苷(5),莫诺苷(6),獐芽菜苷(7),马钱素(8),芍药苷(9),丹皮酚原苷(10),番木鳖酸(11)。结论:该糖苷部位是新药六味地黄苷糖发挥免疫和内分泌调节作用的重要部分。化合物6~8是该部位的主要成分,具有较好的免疫调节作用,是该部位主要活性成分。
OBJECTIVE: To study the chemical constituents of glycosides in Liuwei rehmannia glycosides and elucidate the material basis of its activity. Methods: Glycosidic sites were prepared by macroporous resin column chromatography. The constituents were separated by silica gel and reverse phase silica gel column chromatography. The structures were identified by MS and NMR techniques. Results: Eleven compounds were isolated, including nine iridoid glycosides, one pinane glycoside and one phenolic glycoside, which were identified as 7-dehydrocarbane (1), 7α-methoxy (3), 7α-Ethoxymono-glycoside (4), 7β-Ethoxymono-glycoside (5), Morroniside (6) (7), loganin (8), paeoniflorin (9), paeonol glycoside (10), and loganic acid (11). Conclusion: The glycoside site is an important part of the new drug Liuwei Tuhuang sugar to play an immune and endocrine regulatory role. Compound 6 ~ 8 is the main component of the site, has a good immunomodulatory effect, is the site of the main active ingredient.
