目的:探究EZH2基因在急性髓细胞白血病(AML)细胞表达及其与髓外浸润关系。方法:应用实时荧光定量PCR(qRT-PCR)技术检测42例AML患者骨髓细胞EZH2 mRNA表达,采用免疫组织化学方法检测16例AML患者髓外浸润组织中EZH2蛋白表达水平,并分析EZH2表达与临床特征及预后的关系。结果:42例AML患者骨髓细胞EZH2 mRNA表达水平显著高于正常对照组(P=0.005),EZH2 mRNA表达水平与男性、外周血幼稚细胞比例≥60%、WBC≥35×109/L及发病时伴有髓外浸润显著相关,且与外周血幼稚细胞比例和WBC计数呈显著正相关(P=0.009、P=0.023),另外伴有髓外浸润组EZH2 mRNA表达显著高于无髓外浸润组(P=0.001)。16例AML髓外浸润组织中,10例患者阳性表达EZH2蛋白,且其中7例患者中至强阳性程度表达EZH2。结论:EZH2在AML患者中高表达,可能通过某种机制参与白血病细胞髓外浸润的过程。
Objective: To investigate the expression of EZH2 gene in acute myeloid leukemia (AML) cells and its relationship with extramedullary infiltration. Methods: The expression of EZH2 mRNA in bone marrow cells was detected by real-time quantitative PCR (qRT-PCR) in 42 patients with AML. The expression of EZH2 protein in the extramedullary infiltration tissues of 16 patients with AML was detected by immunohistochemistry. Among 16 cases of AML extramedullary infiltration tissues, 10 cases were positive for EZH2 protein, and 7 of them showed moderate to strong positive expression of EZH2. Conclusion: EZH2 is highly expressed in AML patients and may participate in extramedullary infiltration of leukemia cells through some mechanism.
