目的:探讨体重对实验性糖尿病肾病大鼠尿蛋白检测的影响。方法:清洁级健康雄性SD大鼠72只,随机分为糖尿病组(DM组,n=45)和对照组(NC组,n=27),糖尿病组大鼠一次性腹腔注射STZ 60 mg.kg-1。两组大鼠分别在第4,8,12,16周留取24 h尿液,用放免法检测尿白蛋白,监测体重(BW),同时检测肾脏功能和形态。结果:尿白蛋白/肌酐(UACR)在两组间无差异于;尿白蛋白(UTP),尿白蛋白排泄率(UAER),UTP/BW,UAER/BW,UACR/BW在4个时间点DM组高于NC组(P<0.05);5种尿蛋白不同评价指标与肾脏功能形态指标无相关性。结论:体重对糖尿病大鼠UACR的指标评价影响最明显。5种尿白蛋白的评价指标在糖尿病肾病早期不能动态完全反映肾脏病理损害变化。
Objective: To investigate the effect of body weight on urinary protein in rats with experimental diabetic nephropathy. Methods: Seventy-two healthy male Sprague Dawley rats were randomly divided into diabetic group (DM group, n = 45) and control group (NC group, n = 27). Rats in diabetic group were given intraperitoneal injection of STZ 60 mg · kg -1. Urine albumin was detected by radioimmunoassay and body weight (BW) at the 4th, 8th, 12th and 16th week respectively in both groups. Urine function and morphology were detected at the same time. Urinary albumin (UAC), urinary albumin excretion rate (UAER), UTP / BW, UAER / BW and UACR / BW were not significantly different between the two groups at 4 time points DM group was higher than NC group (P <0.05). There was no correlation between different evaluation indexes of urinary protein and renal function morphology index. Conclusion: The body weight has the most obvious influence on the index evaluation of UACR in diabetic rats. The evaluation indexes of five kinds of urinary albumin can not fully reflect the change of pathological changes of kidney in the early stage of diabetic nephropathy.
