目的:观察安息香对雌性孕妊大鼠血清雌二醇、孕酮以及在体子宫平滑肌的影响。方法:依据阴栓观察法和阴道涂片法确定大鼠孕期;通过酶联免疫吸附分析法(ELISA)测定血清雌二醇(E2)及孕酮(PROG)水平,称重法测定子宫和卵巢指数;采用生物机能系统测定不同浓度安息香对孕妊大鼠在体子宫平滑肌张力、频率和峰面积的影响;并观测安息香1.0g/kg对缩宫素、米索前列醇对子宫平滑肌的干预机制。结果:安息香0.5、1.0g/kg组均能显著增加孕妊大鼠血清中雌二醇、孕酮水平,增加卵巢和子宫指数,并能显著抑制在体子宫平滑肌的张力、频率和峰面积;安息香1.0 g/kg能显著对抗缩宫素和米索前列醇后子宫平滑肌的收缩张力、平均面积、频率;再次加入缩宫素能明显增强安息香后的子宫平滑肌的张力和收缩频率;再次加入米索前列醇,对安息香作用后的子宫平滑肌则无影响。结论:安息香能增加孕妊大鼠血清中雌性激素水平,同时可能通过阻断孕妊大鼠在体子宫平滑肌的前列腺受体和部分阻断缩宫素受体而抑制子宫平滑肌运动。
Objective: To observe the effects of benzoin on serum estradiol, progesterone and uterine smooth muscle in pregnant female rats. Methods: Pregnancy was determined according to the method of vaginal plug observation and vaginal smear. Serum estradiol (E2) and progesterone (PROG) levels were determined by enzyme-linked immunosorbent assay (ELISA), and the uterus and ovary Index; using biofunctional system to determine the effect of different concentrations of benzoin on uterine smooth muscle tension, frequency and peak area in pregnant rats; and observe the intervention mechanism of oxybutynin and misoprostol on uterine smooth muscle . Results: The benzoin 0.5 and 1.0 g / kg groups could significantly increase the serum estradiol and progesterone levels, increase the ovarian and uterine index, and significantly inhibit the tension, frequency and peak area of the uterine smooth muscle in the pregnant rats. Benzoin 1.0 g / kg can significantly antagonize the contractile tension, average area and frequency of uterine smooth muscle after oxytocin and misoprostol; the addition of oxytocin can significantly enhance the tension and contraction frequency of uterine smooth muscle after benzoin; Soprostigmine, on the role of benzoin after the uterine smooth muscle had no effect. CONCLUSION: Benzoin can increase estrogen levels in serum of pregnant rats and inhibit uterine smooth muscle movement by blocking the prostate receptor in the pregnant uterine smooth muscle and partially blocking the receptor of oxytocin.
