目的:采用星点设计-效应面法优化碎米糖化液脱色的工艺条件。方法:以脱色剂活性炭用量、pH值和加热温度为考察因素,以葡萄糖保留率和脱色率为效应值,分别采用多元线性回归和二项式方程拟合,用效应面法优化工艺,制备3批样品并进行验证试验。结果:碎米糖化液活性炭脱色的优化工艺为糖化液加入4%的活性炭后,在pH值3.5~4.5、60℃下脱色,3批样品保留率和脱色率预测值与实测值的平均偏差分别为(5.16±0.12)%、(7.13±0.21)%。结论:本文所建立的脱色工艺简便可行,可为进一步制备注射用葡萄糖的脱色工艺研究提供试验依据。
OBJECTIVE: To optimize the decolorization conditions of broken rice saccharification liquid by the method of star point design and response surface. Methods: The amount of activated carbon, pH value and heating temperature of decoloring agent were taken as the investigation factors. The effect of glucose retention and decolorization rate were taken as the effect values. Multivariate linear regression and binomial equation were used respectively. Batch samples and verify the test. Results: The optimal technology for decolorization of activated carbon from broken rice saccharified liquid was as follows: after adding 4% activated carbon into the saccharified solution, decolorizing at pH 3.5-4.5 and 60 ℃, the average deviation between predicted and measured value of retention and decolorization rate (5.16 ± 0.12)%, (7.13 ± 0.21)%. Conclusion: The decolorization process established in this paper is simple and feasible, which can provide experimental basis for the further study on the decolorization process of glucose for injection.
