目的:探讨体外细胞培养,观察核转录因子κB通路特异性抑制剂吡咯烷二硫代氨基甲酸酯(PDTC)对膀胱癌EJ细胞凋亡抑制蛋白Livin、Survivin mRNA表达水平的影响。方法:采用Real-time PCR法检测以不同浓度不同时间的PDTC处理的膀胱癌EJ细胞的凋亡抑制蛋白Livin、Survivin mRNA表达水平情况。结果:PDTC可降低膀胱癌EJ细胞凋亡抑制因子Livin、Survivin mRNA表达水平,呈剂量-时间依赖性,且呈负相关(P<0.05)。结论:PDTC可使膀胱癌EJ细胞凋亡抑制蛋白Livin、Survivin mRNA表达水平减少,其机制可能与NF-κB通路被抑制相关。
OBJECTIVE: To investigate the effect of pyrrolidine dithiocarbamate (PDTC), a specific inhibitor of nuclear factor κB (NF-κB), on the expression of Livin and Survivin mRNA in bladder cancer EJ cells in vitro. Methods: Real-time PCR was used to detect the expression of Livin and Survivin mRNA in bladder cancer EJ cells treated with different concentrations of PDTC. Results: PDTC decreased the expression of Livin and Survivin mRNA in bladder cancer EJ cells in a time-and dose-dependent manner (P <0.05). Conclusion: PDTC can reduce the expression of Livin and Survivin mRNA in bladder cancer EJ cells, which may be related to the inhibition of NF-κB pathway.
