目的:明确增重剂对茜草饮片质量及其所含大叶茜草素和羟基茜草素含量的影响。方法:采用高效液相色谱法,色谱柱为资生堂Tech Mate CACELL PAK C18柱(250mm×4.6mm,5μm),流动相为乙腈-0.1%三氟乙酸,流速:1.0m L/min,检测波长:240nm,柱温:30℃。结果:大叶茜草素在10-500μg/m L范围内线性良好,羟基茜草素在5-200μg/m L范围内线性良好;两者的平均加样回收率分别为99.32%(RSD=2.26%)和99.59%(RSD=2.63%);实验组茜草的质量均不低于对照组茜草,且对照组茜草中羟基茜草素和大叶茜草素的含量均明显高于实验组。结论:增重剂可增加茜草的质量并明显降低茜草中羟基茜草素和大叶茜草素的含量,同时还通过改变色谱图中各峰的比例影响茜草的质量。
Objective: To clarify the effect of weighting agent on the quality of Rubia cordifolia pieces and the content of phleum carporib and phorbolide contained in it. Methods: High performance liquid chromatography (HPLC) was performed on a Shiseido Tech Mate CACELL PAK C18 column (250 mm × 4.6 mm, 5 μm) with a mobile phase of acetonitrile 0.1% trifluoroacetic acid at a flow rate of 1.0 mL / 240nm, column temperature: 30 ℃. Results: The linear regression equation was established in the range of 10-500 μg / m L and the linear calibration curve was found in the range of 5-200 μg / m L. The average recoveries of the two compounds were 99.32% (RSD = 2.26% ) And 99.59% (RSD = 2.63%) respectively. The quality of Rubia cordifolia in the experimental group was not lower than that in the control group, and the contents of both cademy and cauliflower in the control group were significantly higher than those in the experimental group. CONCLUSION: The weight-increasing agent can increase the quality of Rubia cordifolia and significantly reduce the content of Rubiacetin and Rubiacetin in Rubia cordifolia, and also influence the Rubia cordifolia by changing the ratio of each peak in the chromatogram.
