提要:目的观察缺血再灌注大鼠肠上皮细胞mtDNA ATPase 6,8亚基基因的改变情况。方法将6只Wistar大鼠分成对照组、缺血再灌注组。分别提取mtDNA,采用PCR技术扩增后测序,检测突变点。结果Wistar大鼠mtDNA ATPase 6,8亚基基因存在多态性,在缺血再灌注组存在a7 797缺失和一定数量点突变:A7 779T,T8 515G,T8 520G,C8 535G。结论缺血再灌注后,肠上皮细胞mtDNA ATPase 6,8亚基基因存在损伤,由此可能导致所编码的氨基酸改变。
Abstract: Objective To observe the changes of mtDNA ATPase 6 and 8 subunit genes in intestinal epithelial cells during ischemia-reperfusion in rats. Methods Six Wistar rats were divided into control group, ischemia-reperfusion group. The mtDNAs were extracted separately and amplified by PCR and sequenced to detect the mutation points. Results The mtDNA ATPase 6 and 8 subunit genes of Wistar rats were polymorphic. There was a7797 deletion and a certain number of point mutations in the ischemia / reperfusion group: A7 779T, T8 515G, T8 520G and C8 535G. Conclusion After ischemia-reperfusion, the mtDNA ATPase 6,8 subunit gene of intestinal epithelial cells is damaged, which may result in the change of amino acid encoded.
