目的:观察淫羊藿苷和固本方对肾虚型老年痴呆模型大鼠空间学习记忆能力及海马Aβ表达的影响。方法:选用SD大鼠,随机分为对照组、模型组、淫羊藿苷组,固本方组。采用皮下注射氢化可的松联合侧脑室注射Aβ25-35建立肾虚型老年痴呆大鼠模型。造模后,给药4周。结果:模型组皮质酮及睾酮含量明显低于空白组(P<0.05),结合Morris水迷宫测试,提示肾虚型老年痴呆大鼠模型复制成功;淫羊藿苷组和固本方组较模型组第1次抵达原平台时间缩短(P<0.01);与固本方组比较,淫羊藿苷发现平台时间未见明显异常;dot blots检测结果,模型组Aβ表达强于空白组(P<0.01);淫羊藿苷组和固本方组Aβ的表达明显减少(P<0.05,P<0.01)。结论:淫羊藿苷和固本方组可改善肾虚型老年痴呆模型大鼠空间学习记忆能力,并可降低海马内Aβ的表达。
Objective: To observe the effects of icariin and Gubenfang on spatial learning and memory and the expression of Aβ in hippocampus of Alzheimer’s disease model rats with kidney deficiency. Methods: SD rats were randomly divided into control group, model group, icariin group and Gubenfang group. Subcutaneous injection of hydrocortisone combined with intracerebroventricular injection of Aβ25-35 to establish kidney-type dementia rat model. After modeling, given for 4 weeks. Results: The levels of corticosterone and testosterone in the model group were significantly lower than those in the blank group (P <0.05). Combined with the Morris water maze test, the model of Alzheimer’s disease rat model was successfully replicated. Compared with the model group Compared with Gubenfang group, Icariin showed no obvious abnormality in platform time. The results of dot blots showed that the expression of Aβ in model group was stronger than that in blank group (P <0.01) ). The expression of Aβ in icariin group and Gubenfang group decreased significantly (P <0.05, P <0.01). Conclusion: Icariin and Gubenfang group can improve spatial learning and memory abilities in Alzheimer ’s disease model rats and reduce Aβ expression in hippocampus.
