目的:探讨H2S对巨噬细胞COX-2表达及泡沫化的影响。方法:建立氧化低密度脂蛋白(Ox-LDL)诱导RAW264.7细胞泡沫化模型,同时给予不同浓度的H2S。应用油红O染色观察泡沫细胞的形成;Real-time qPCR检测COX-2 mRNA的表达;Western blot检测COX-2蛋白的表达。结果:Ox-LDL能够明显增加细胞内脂质的沉积,同时伴有COX-2的产生。不同浓度的H2S可以明显减少由Ox-LDL引起的COX-2的表达,同时也减少了细胞内脂质的沉积,以250μmol/L最为明显,差异具有统计学意义(P<0.05)。结论:H2S可以显著减少巨噬细胞COX-2的表达及泡沫化。
Objective: To investigate the effect of H2S on COX-2 expression and foam formation in macrophages. Methods: Oxidized low-density lipoprotein (Ox-LDL) was used to induce RAW264.7 cells foaming model with different concentrations of H2S. The formation of foam cells was observed by oil red O staining. The expression of COX-2 mRNA was detected by Real-time qPCR. The expression of COX-2 protein was detected by Western blot. RESULTS: Ox-LDL significantly increased intracellular lipid deposition accompanied by COX-2 production. Different concentrations of H2S can significantly reduce the expression of COX-2 induced by Ox-LDL, and also reduce the deposition of intracellular lipid, with 250μmol / L being the most obvious, the difference was statistically significant (P <0.05). Conclusion: H2S can significantly reduce the expression of COX-2 macrophages and foam.
