目的探讨单核细胞趋化蛋白-1(MCP-1)及其受体CC趋化因子受体2(CC chemokine receptor 2.CCR2)在小鼠阴道念珠菌病发病中的作用。方法建立阴道念珠菌病小鼠模型.以RT-PCR及ELISA方法检测小鼠阴道及腰淋巴结中MCP-1及CCR2的表达。结果与正常小鼠相比,雌激素对MCP-1及CCR2的表达无明显影响。所有感染组小鼠阴道组织MCP-1的mRNA和蛋白水平均明显升高,其CCR2 mRNA水平亦相应升高;与之相反,所有感染组小鼠的引流腰淋巴结MCP-1的浓度与基础水平相比无明显改变,而雌激素处理感染组小鼠CCR2在接种后7~21d明显升高。结论MCP-1及其受体CCR2可能在局部刚道黏膜白念珠菌感染中起一定的作用。
Objective To investigate the role of monocyte chemoattractant protein-1 (MCP-1) and its receptor CC chemokine receptor 2 (CCCR2) in the pathogenesis of vaginal candidiasis in mice. Methods To establish a vaginal candidiasis mouse model. The expression of MCP-1 and CCR2 in vaginal and lumbar lymph nodes was detected by RT-PCR and ELISA. Results Compared with normal mice, estrogen had no significant effect on the expression of MCP-1 and CCR2. The mRNA and protein levels of MCP-1 in the vaginal tissues of all infected mice were significantly increased, and the CCR2 mRNA levels were also increased accordingly. In contrast, the concentrations of MCP-1 in the lumbar lymph nodes of all the infected mice were similar to those of the basal level Compared with no significant change, while estrogen-treated mice CCR2 significantly increased 7 ~ 21d after inoculation. Conclusion MCP-1 and its receptor CCR2 may play a role in the infection of local pathogenic Candida albicans.
