目的:优选蜣螂提取物的DA201-C型大孔树脂纯化工艺。方法:采用单因素试验考察DA201-C型树脂的吸附性能和影响纯化工艺的pH、洗脱剂、上柱流速等因素;采用氨基酸自动分析仪检测纯化后的中间体。结果:最佳纯化工艺条件为上样液质量浓度17.60 g.L-1,吸附流速2 BV.h-1,pH 4.0,依次用1 BV水和4 BV 70%乙醇洗脱,脱盐率达98.81%,多肽回收率达87.39%,所得醇洗脱物含结合型氨基酸(多肽)32.26%。结论:该工艺分离效果较好,可用于蜣螂中间体的脱盐与纯化。
Objective: To optimize the DA201-C macroporous resin purification technology of dung beetle extract. Methods: The single factor test was used to investigate the adsorption capacity of DA201-C resin and the factors influencing the pH value, eluent and column flow rate of the purification process. The purified intermediates were detected by automatic amino acid analyzer. Results: The optimal purification conditions were as follows: the mass concentration of the sample was 17.60 gL-1, the adsorption flow rate was 2 BV.h-1, the pH was 4.0. The elution rate was 98.81% with 1 BV water and 4 BV 70% Polypeptide recovery rate of 87.39%, the resulting alcohol eluate containing binding amino acids (polypeptides) 32.26%. Conclusion: The separation of the technology is better, can be used for the desalination and purification of dung beetle intermediates.
