目的:探讨8-硝基白杨素有无化疗增敏的作用及诱导细胞凋亡的原理。方法:体外培养卵巢癌HO-8910细胞,经8-硝基白杨素和顺铂处理后,以3-(4,5)-双甲基-2-噻唑-(2,5)-二苯基溴四氮唑蓝(MTT)法检测细胞的增殖情况;以流式细胞仪检测细胞凋亡。结果:8-硝基白杨素联合顺铂合用72 h时抑制率高达71.8%,与顺铂单独作用相比差异有统计学意义(P<0.05)。两药合用时抑制率达到最高峰。结论:体外实验中,顺铂可抑制卵巢癌HO-8910细胞增殖并诱导其发生凋亡,且与8-硝基白杨素联合应用时抑制作用明显增加。
Objective: To investigate the effect of 8-nitrocyanin on the chemosensitization and the principle of apoptosis induction. METHODS: HO-8910 cells were cultured in vitro and treated with 8-nitrocyanin and cisplatin. The cells were treated with 3- (4,5) -dimethyl-2-thiazolyl- Broth (MTT) method was used to detect cell proliferation; apoptosis was detected by flow cytometry. Results: The inhibition rate of 72-h combined with cisplatin and 8-nitrocyanin was as high as 71.8%, which was significantly different from that of cisplatin alone (P <0.05). The two drugs combined inhibition rate reached its peak. CONCLUSIONS: In vitro, cisplatin inhibits the proliferation and induces apoptosis of HO-8910 cells, and its inhibitory effect is obviously increased when combined with 8-nitrocapsin.
