MicroRNAs(miRNAs)是一类长度约为19~25 nt内源性的、非编码单链小RNA,在植物逆境胁迫应答中具有重要的调节作用。为了获得棉花中黄萎病抗性相关的miRNA,利用生物信息学技术,本研究从Sanger miRNA数据库和已报道的文献中找到64个与棉花黄萎病抗性相关的miRNA,预测和分析了它们的靶基因和启动子,并从中筛选出12个miRNA进行了实时荧光定量PCR分析,为进一步深入研究miRNA的作用机制奠定基础和提高转基因棉花抵抗黄萎病的能力提供新的途径。
MicroRNAs (miRNAs) are a class of small, endogenous, non-coding, single-stranded small RNAs of about 19-25 nt in length that have important regulatory roles in plant stress response. In order to obtain miRNAs related to Verticillium wilt resistance in cotton, 64 biomarkers of miRNAs resistant to Verticillium wilt were obtained from the Sanger miRNA database and reported papers using bioinformatics techniques. Of the target genes and promoters, and 12 miRNAs were screened out for real-time fluorescence quantitative PCR analysis, providing a new way for further study of the mechanism of action of miRNA and improve the ability of transgenic cotton to resist Verticillium wilt.
