目的:探讨健骨颗粒含药血清对体外培养大鼠成骨细胞增殖的影响。方法:采用酶消化法培养SD大鼠成骨细胞,健骨颗粒含药血清干预,以生理盐水大鼠血清为对照,运用MTT法、流式细胞术检测成骨细胞增殖情况及细胞周期分布。结果:与同浓度生理盐水血清相比,20%健骨颗粒含药血清能明显促进体外培养成骨细胞增殖(P<0.05),明显降低G0/G1期细胞分数(P<0.01或P<0.05),提高S期、G2/M期细胞分数和增殖指数(P<0.01或P<0.05)。结论:健骨颗粒含药血清能推进体外培养成骨细胞细胞周期,从而促进成骨细胞增殖。
Objective: To investigate the effect of Jiangu granule-containing serum on proliferation of rat osteoblasts in vitro. Methods: SD rat osteoblasts were cultured by enzymatic digestion. Serum containing Jiangu Granules was used for intervention. Serum of normal saline rats was used as control. The proliferation and cell cycle distribution of osteoblasts were detected by MTT assay and flow cytometry. Results: Compared with the same concentration of saline serum, 20% Jiangu granules serum could significantly promote the proliferation of cultured osteoblasts in vitro (P <0.05) and significantly decrease the cell fraction in G0 / G1 phase (P <0.01 or P <0.05 ), And increased the cell fraction and proliferation index in S phase and G2 / M phase (P <0.01 or P <0.05). CONCLUSION: Jiangu granule-containing serum can promote the cell cycle of osteoblasts in vitro and promote the proliferation of osteoblasts.
