目的:探讨高质量蛋白抗体的制备,为蛋白质的研究提供有效的工具。方法:用人绒毛膜促性腺激素(h CG)免疫BAL B/ c小鼠制备其单克隆抗体(Mb) ;以p ICln蛋白的18肽为抗原对兔进行免疫注射制备血清抗体,用EL ISA分别检测抗h CG的单克隆抗体腹水和p ICln蛋白的肽段血清抗体的效价及特异性。结果:肽段血清抗体在12 80 0倍稀释时也呈阳性反应且高特异性。结论:从制备方法的复杂程度、抗体的效价及特异性等方面看,肽段血清抗体的制备是一种方便有效地制备蛋白质研究用抗体的方法。
Objective: To investigate the preparation of high quality protein antibody and provide an effective tool for the study of protein. METHODS: BALB / c mice were immunized with human hCG (hCG) to prepare their monoclonal antibodies (Mb). Serum antibodies were prepared by immunization of rabbits with 18 peptide of pIcln as antigen and ELISA Titers and specificities of sera from patients with anti-hCG monoclonal antibody ascites and pIcln serum were detected. RESULTS: Peptide serum antibodies were also positive and highly specific at 12 80-fold dilutions. CONCLUSION: The preparation of peptide serum antibody is a convenient and effective method for the preparation of protein research antibodies in terms of the complexity of the preparation method, the potency and specificity of the antibody, and the like.
